Shiki Misaki had led a relatively normal life for her first fifteen years. She had grown up with regular parents, and gone to school just like everyone else. She aspired to be a fashion designer, and together with her best friend Eri, she designed and created clothing on a frequent basis, to the point where nearly everything she wore was of her and Eri's own design and creation. Throughout this time, however, Shiki was consumed with an intense jealousy of Eri - how cute she was, popular, talented, etc. - and, as much as she was her friend, she also wanted to be her just as badly.
When she was fifteen however, there was a day when she and Eri got into a fight. Eri told her that she wasn't cut out to be a designer, Shiki stormed out, and subsequently got into an accident involving a bus and died.
Upon dying, Shiki woke up and found herself in the Reaper's Game. The basic idea of the Reaper's Game is to survive seven days, completing various missions each day, fighting monsters called Noise, and trying not to be Erased which would lead to True Death, or oblivion. If, however, she could survive those seven days and win the game, she would be allowed to come back to life.
In the Game, you need a partner to survive, and she formed a Pact with another Player named Neku Sakuraba. Their relationship was tenuous at best in the beginning. She thought that he was constantly running off and ignoring her, and he thought that she was basically stalking him. In fact, on the Second Day, Neku was tricked into attempting to Erase her, and nearly went through with it before he was stopped by another person- Mr. Sanae Hanekoma, who was able to save both Shiki and Neku, and help them start learning how to trust each other and work together to win.
One essential aspect of the Game is that you have to pay an entry fee to be a Player - not unlike a character's main loss in Paradisa. Shiki's loss was her body. She woke up in the Game, and found herself looking exactly like Eri. And when she saw herself, she freaked, but in a good way. She was incredibly happy, thought that she could finally be exactly the way she wanted, and vowed to be more bubbly and cheerful - just like Eri - so she could be the person she wanted to be.
Throughout the course of the week, however, several things happened. Her jealous tendancies were revealed to Neku, and he began to help her work through them, so she could be happy with who she was on her own. She managed to show him the value of friendship and caring about others, and help him come out of his shell a little. They worked together, fought together, and by the end of the week, they had grown to be real friends who genuinely cared for and trusted each other.
They won the Game. But then they hit a snag. Theoretically, when Players win the Reaper's Game, they are allowed to come back to life. However, when Neku and Shiki won, they faced the Conductor of the Game, Megumi Kitaniji, and he informed them that only one of them would be allowed to come back to life. Shiki would be allowed to come back. Neku was offered the choice between Erasure and playing the Game a second time. Shiki offered to trade places with him, but he declined, and Kitaniji refused to allow it anyway, so Neku entered the Game again, and Shiki was supposed to come back to the Real Ground (also known as the RG, or our layer of the world... the Reaper's Game takes place in Shibuya, which is where all of the characters originally were, but in an alternate version of it, known as the UG, or Underground).
The next two weeks for Shiki are a blank, because she did not, in fact, come back to life. In those next two weeks, during which Neku played the Game twice, his Entry Fee was changed to be her, so she was held in limbo as he played for her.
She next comes to conciousness in the final day of Neku's third and final Game. Neku and his then-partner, Daisukenojo "Beat" Bito, are trying to find the head of the Reaper's Game, also known as the Composer, to defeat him and make Beat the Composer in his place. Shiki doesn't entirely understand what's going on, but she goes along with them, trusting Neku implicitly, to aid their fight.
Their first opponent is the Conductor, Kitaniji, and Shiki is all set to aid Neku and Beat against him, but she then goes blank again - Kitaniji takes over her mind, and forces her body to fight against her friends. She later comes to with Beat waking her up after Neku had gone on ahead to find and face the Composer, all after they had beaten Kitaniji and subdued herself. The pair of them then go after Neku and help him fight, and beat, the Composer.
After this final battle, they are all finally allowed to come back to life and be regular people again. They are all irrevocably changed however, all being incredibly close friends now, to the point where they all know Shiki even when they finally get to see her for the first time in her own body, and all having incredibly strong Imaginations and fighting skills.
And then, for Shiki, it is shortly after this, when her life is finally returning to normal and she is back in her own body again, when she gets whisked away to Paradisa.
Personality

Bright, bubbly, and cheerful, Shiki Misaki is typically a little ray of sunshine who enjoys making people happy and just having fun. At least, that's the way she appears on the outside. Inside, however, she's much more bitter than that. Not to the point where it's debilitating and it messes with her everyday life, but she used to suffer from intense jealousy of one of her best friends, and even as she's getting over that now, it's still something that she struggles with every day.
That jealousy stems from her own insecurity. Not only does she have the massive self-esteem issues that any normal 15-year-old girl has, but her jealousy makes her think even less of herself. This is something that she tends to hide from others, but every compliment someone pays means the world to her... and she's usually fairly quick to jump to the conclusion that she's done something wrong.
All of this, however, is almost always hid behind her facade of cheerfulness, happiness, a touch of snark when it's appropriate, and constant bubbliness and self-confidence. You need to actually know her to see what's going on beneath.
Canon relationships

Neku Sakuraba - Neku is her Partner and grows to be her best friend, despite their rocky beginning. She loves him and trusts him completely.
Raimu "Rhyme" Bito - Rhyme is special to Shiki as she's the other female player she knows in the Game. She doesn't get to know her that well in canon as Rhyme gets Erased halfway through the first week, but they hit it off instantly, and Shiki would still consider her to be a good friend.
Daisukenojo "Beat" Bito - Beat is someone Shiki constantly has to care and look out for, especially after Rhyme is Erased, but she still considers him to be a good friend, and she trusts him quite a lot.
Yoshiya "Joshua" Kiryu - She doesn't know much about him since she doesn't meet him in person until the end of her time in the Reaper's Game. She knows that he was Neku's second Partner, so she trusts him because Neku trusts him (to an extent), but she only met him as the Composer of the Reaper's Game, and someone she had to fight, so she's still wary around him.
Sanae Hanekoma - Almost a mentor to her and Neku in the Game, Mr. Hanekoma is the one who saves Neku from Erasing her, and gives them advice and help along the way. Why he was there with Joshua at the end, after she, Neku and Beat beat him is beyond her, but she still respects and trusts him.
Megumi Kitaniji - Despite him watching over her for a week, and later controlling her mind, she doesn't know a lot about this man. All she knows is that he was the Conductor of the Reaper's Game, that he's the one who denied Neku from coming back to life the first time, and that he's the one who took over her brain and made her fight her friends later on. She's incredibly wary of him, distrusts him... but luckily, does not have much to do with him.
Koki Kariya - She doesn't have much to do with him either. He's a Reaper, but he's fairly chill for a Reaper... doesn't particularly care for doing work, so while she knows that he will fight if he has to, she never made him have to.
Uzuki Yashiro - She's the one who nearly tricked Neku into Erasing her. She doesn't like her.
Yodai Higashizawa - He was the Game Master the week that she was actively Playing the Game. He was the one who assigned missions, and he was the major boss she and Neku had to fight at the end of their week. On top of that, he has truly horrendous fashion sense - he wears a ram on his crotch, for instance. He is another person that Shiki tends to steer clear of.

So far, she has only had to deal with her main loss in Paradisa, the loss of her body, so she looks like Eri again. She has had to cope with various plots within the castle - most notably the babies plot where she completely fell in love with the child the castle gave her - but the only real loss she has had thus far has been her main one.
Relationships

Equius Zahhak - In human terms, he's her boyfriend. In troll terms, she is his matesprit. Either way, they are romantically together, and this is something that is new but awesome for her.
Maya Fey - She has known her for a fair amount of the time that she has been in Paradisa, and she would say that Maya's a friend. They're not super close, but she really enjoys her company, and would always love to spend more time with her.
Barney Stinson - He started out as someone who just knew more about Paradisa than she did, but they kept talking every so often... and now he's her boss at the in-Paradisa WildKat, where she works. She doesn't know him all that well either, but she enjoys his company.
Ted Mosby - He's pretty new to Paradisa, from Shiki's point of view, but they've been talking a fair amount, and she'd like to keep talking to him even more!
Draco Malfoy - A pretty good friend of hers in Paradisa, even if she's had to deal with him through various losses and plots that made the both of them do strange things, he's usually been there to explain some basic things to her, and she really appreciates having him as a friend.
Suoh Tamaki - Her regular host at the Host Club, and a good friend of hers. Knowing him is one of the things that makes her not totally hate being in the castle, and a conversation with him is always guaranteed to make her feel better... he's someone she really cares about, and she wants to be there for him just as much as he's been there for her when she's needed him, if not more.
Mitsukuni "Hunny" Haninozuka - OH MY GOD HE'S SO CUTE. No, seriously, she absolutely loves Hunny for how adorable and sweet he is. They've had their issues in the past - such as a loss on his end that made him go all "dark Hunny" on her that really hurt - but they're past that now, and she really enjoys being around him.
Ritsu Kasanoda - It's been a while since she's talked to him, but she likes spending time with him, and figuring out puzzles and riddles with him, and she would love to see him again at some point soon!
Nepeta Leijon - Shiki doesn't know her all that well. Really, they kind of just met. But she's Equius's moirail, so Shiki supposes that she'll get to know her better soon... which isn't really all that much of a problem by her! She's enjoyed the conversation that she's had with her, thus far.
